«Οὐκ ἀγνοεῖ γὰρ ὅτι ἀπὸ τοσούτων καρπῶν μέλλει πολὺ ἔλαιον ποιεῖν.»

This just doesn't make sense to me; especially the «πολὺ ἔλαιον» at the end.

My best guess would be, "For he doesn't know what he is going to do [with] a lot of olive oil from so much fruit."

Can an accusative be used like that? Or am I just totally off?

It's just that ποιῶ covers the meanings of both "make" and "do", and here the first is what's meant. That should clear that up.

Another thing is that you have to be careful with ἀγνοῶ as it means "to not know", so οὐκ ἀγνοεῖ means "he is not ignorant". And ὅτι is "that" rather than "what".
